13D filed on July 10, 2019, as amended by Amendment No. 1 to Schedule 13D filed on June 1, 2020 and Amendment No. 2 to Schedule 13D filed on October 6, 2020 (as so amended, the “Schedule 13D”). Except as specifically provided herein, this Amendment
No. 3 does not modify any of the information previously reported in the Schedule 13D. Unless otherwise indicated herein, capitalized terms used but not defined in this Amendment No. 3 shall have the same meanings herein as are ascribed to such terms

Item 4.

Purpose of Transaction.
|
Item 4 of the Schedule 13D is hereby amended and supplemented as follows:
On February 11, 2021, KKR Genetic Disorder L.P., as selling stockholder, and the Issuer entered into an underwriting agreement (the “Underwriting Agreement”) with Goldman Sachs & Co. LLC, as representative for the underwriters party thereto
(the “Underwriters”), providing for the offer and sale of 3,000,000 shares of Common Stock by KKR Genetic Disorder L.P. (the “Offering”), and purchase by the Underwriters of the shares of Common Stock at a price of $62.50 per share of Common Stock.
Pursuant to the Underwriting Agreement, KKR Genetic Disorder L.P. also granted to the Underwriters a 30-day option to purchase up to an additional 450,000 shares of Common Stock, which was exercised in full on February 12, 2021. The sale of 3,450,000
shares in the Offering closed on February 17, 2021. The Offering was made pursuant to the Issuer’s shelf registration statement on Form S-3 (File No. 333-240147), as supplemented by a prospectus supplement dated February 11, 2021.
Pursuant to the Underwriting Agreement, KKR Genetic Disorder L.P. has entered into a lock-up agreement (the “Lock-Up Agreement”) with the Underwriter pursuant to which it has agreed with the Underwriter, subject to customary exceptions, not to
offer for sale, sell, pledge, or otherwise dispose of (or enter into any transaction or device that is designed to, or could be expected to, result in the disposition by any person at any time in the future of) any shares of Common Stock or
securities convertible into or exercisable or exchangeable for Common Stock during the period from February 11, 2021 continuing through the date 60 days thereafter, except with the prior written consent of the Underwriter. The foregoing description
of the Lock-Up Agreement set forth in this Item 4 does not purport to be complete and is qualified in its entirety by reference to the full text of the Lock-Up Agreement, which has been filed as Exhibit H hereto and is incorporated herein by
reference.

& Co. (as the sole shareholder of KKR Group Holdings); KKR Management (as the Series I preferred stockholder of KKR & Co.); and Messrs. Kravis and Roberts (as the founding partners of KKR Management) may be deemed to be the beneficial owner
of the securities held directly by KKR Genetic Disorder, in each case, as described more fully in this Schedule 13D.
